A novel CRISPR screening platform maps live cell-extrinsic phenotypes to CRISPR perturbations at high throughput

This research presents a CRISPR screening workflow that maps cell-extrinsic immune phenotypes to specific genetic perturbations at high throughput. Using CellCage enclosures, single edited dendritic cells are paired with T cells, allowing longitudinal imaging of activation and proliferation before matched sgRNA and transcript capture. The study shows how individual perturbations, such as β2M knockout, can be directly linked to changes in antigen presentation and T-cell priming capacity. By resolving function at the level of single cell-cell interactions, the platform captures immune behavior that would be obscured in pooled or endpoint assays. This creates a powerful new framework for discovering regulators of dendritic cell function in immunosuppressive settings.
