This poster presents a multimodal workflow for identifying drug-resistant clones based on both live-cell behavior and transcriptomic state. Using CellCage enclosures, individual lung cancer cells are tracked during treatment with EGFR inhibitors, then linked to matched sequencing data from the same cells. The study reveals distinct resistant phenotypes, including continued proliferation, survival with cell-cycle arrest, and daughter cells that resist death. These dynamic behaviors correspond to different gene expression programs and pathway signatures that would be difficult to infer from endpoint assays alone. The workflow offers a powerful strategy for understanding treatment response and resistance in heterogeneous cancer cell populations.
