Longitudinal Multi-Modal Single-Cell Profiling Reveals Functional Heterogeneity In Human Preadipocytes

This study uses the Cellanome R3200 and CellCage enclosures to track lipid droplet accumulation and viability in individual differentiating preadipocytes alongside matched single-cell transcriptomics. The longitudinal, multimodal approach reveals substantial asynchrony in adipogenic timing and identifies 227 genes, including CCND1 and AKR1B1, that predict lipid accumulation even within transcriptomically similar cell states. By resolving functional heterogeneity invisible to transcriptomics alone, the work offers a new framework for studying stem cell fate decisions relevant to obesity and metabolic disease.
