This poster describes a workflow for profiling T-cell functional heterogeneity in response to checkpoint inhibitors using longitudinal, multimodal single-cell analysis. By monitoring live-cell behavior within CellCage enclosures and linking those phenotypes to downstream molecular data, the platform captures variation in activation, cytotoxicity, and treatment response across individual cells. The approach is designed to reveal functional subpopulations that may be obscured by endpoint or bulk assays. Preserving temporal context makes it possible to distinguish transient from sustained response states. This framework offers a promising path for improving how immunotherapy responses are characterized and compared.
